目的 探讨Caspase -2和caspase -3蛋白在顺铂诱导卵巢癌细胞系A2 780和A2 780 /DDP凋亡中的作用和对A2 780 /DDP细胞系顺铂耐药形成的可能影响。方法 用细胞培养方法 ,加入不同浓度 (分别为 0、5、10、2 0、40 μM)顺铂共同培养卵巢癌细胞系A2 780和A2 780 /DDP 2 4h、48h和 72h ,原位末端标记法 (TUNEL)检测卵巢癌细胞系凋亡情况 ,用免疫组化SABC法观察此过程中Caspase -2和caspase -3蛋白表达。结果 随顺铂作用时间延长和剂量增加 ,卵巢癌细胞系A2 780和A2 780 /DDP中凋亡逐渐增强(P <0 0 1) ,caspase -2和caspase -3蛋白表达呈现出对顺铂作用时间和剂量的依从性 (P <0 0 1) ,两种蛋白在A2 780细胞中表达明显强于A2 780 /DDP( P <0 0 1)。结论 Caspase -2和caspase -3蛋白活化促进顺铂诱导的卵巢癌细胞系A2 780和A2 780 /DDP凋亡 ,其表达抑制可能是影响顺铂耐药的A2 780 /DDP细胞凋亡的原因。
Objective To investigate the role of caspase-2 and caspase-3 in the apoptosis of ovarian cancer cell lines A2 780 and A2 780 / DDP induced by cisplatin and the possible effect on the formation of cisplatin resistance in A2 780 / DDP cell line. Methods Ovarian cancer cell lines A2807 and A2805 / DDP were cultured with different concentrations of cisplatin (0, 5, 10, 20, 40 μM, respectively) for 4 h, 48 h and 72 h with cell culture. (TUNEL) was used to detect the apoptosis of ovarian cancer cell lines. The expression of Caspase-2 and caspase-3 in this process was observed by immunohistochemical SABC method. Results The apoptosis of ovarian cancer cell lines A2807 and A2807 / DDP gradually increased (P <0.01), and the expression of caspase-2 and caspase-3 protein showed cisplatin effect Time and dose compliance (P <0.01). The expression of both proteins in A2 780 cells was significantly stronger than that in A2 780 / DDP cells (P <0.01). Conclusion The activation of caspase - 2 and caspase - 3 can promote the apoptosis of ovarian cancer cell lines A2807 and A2805 / DDP induced by cisplatin. The inhibition of the expression of caspase - 2 and caspase - 3 might be the reason of the apoptosis of A2807 / DDP cells.
