目的利用荧光原位杂交技术(FISH)检测宫颈病变患者宫颈脱落细胞中TERC基因,评价该技术诊断价值。方法选择2007年12月至2009年4月宫颈上皮内瘤变(CIN)患者91例和健康对照者20例,利用FISH技术检测宫颈脱落细胞TERC基因,计数不平衡扩增细胞,比较其在各级宫颈病变中的表达差异。结果对照组、CINⅠ/湿疣、CINⅡ、CINⅢ及以上患者的宫颈脱落细胞中,每100个细胞中TERC基因不平衡扩增细胞分别为(0.1±0.3)个、(0.6±1.3)个、(1.7±2.1)个、(2.5±4.4)个。CINⅡ及以上病变组TERC基因不平衡扩增与对照组和CINⅠ/湿疣组有显著性差异(P=0.018,P=0.002)。结论随宫颈病变加重,TERC基因不平衡扩增细胞增加,FISH技术检测宫颈脱落细胞TERC基因扩增可以辅助诊断宫颈癌前病变。
Objective To detect TERC gene in cervical exfoliated cells of cervical lesions by fluorescence in situ hybridization (FISH), and to evaluate the diagnostic value of this technique. Methods 91 patients with cervical intraepithelial neoplasia (CIN) from December 2007 to April 2009 and 20 healthy controls were enrolled in this study. The TERC gene was detected by FISH and the unbalanced cells were counted. Grade cervical lesions in the expression differences. Results The unbalanced TERC expansion cells per 100 cells in control group, CINⅠ / genital warts, CINⅡ, CINⅢ and above patients were (0.1 ± 0.3), (0.6 ± 1.3), ± 2.1), (2.5 ± 4.4). The unbalanced amplification of TERC gene in CINⅡ and above lesions was significantly different from that in control group and CINⅠ / genital warts (P = 0.018, P = 0.002). Conclusion With the increase of cervical lesions, imbalanced TERC gene amplification cells increased, FISH detection of cervical exfoliated cells TERC gene amplification can help diagnose cervical precancerous lesions.
