目的 :分析可刺激人γδT细胞增殖的结核杆菌多肽抗原(Mtb Ag)的某些生物学特性。方法 :以结核杆菌分泌蛋白与经透析或链霉蛋白酶处理的Mtb Ag体外分别刺激人外周血单个核细胞 (PBMC)增殖 ,培养 10d时 ,采用流式细胞仪分析细胞的表型。结果 :透析后的Mtb Ag刺激γδT细胞增殖的活性未见明显降低 ;而经链霉蛋白酶处理的Mtb Ag促进γδT细胞增殖的活性则显著下降。另外 ,与结核杆菌分泌蛋白相比较 ,Mtb Ag刺激γδT细胞增殖的活性显著增强。 结论 :Mtb Ag中可刺激人γδT细胞增殖的成分是一种相对分子质量 (Mr)大于 10 0 0 0、非分泌性、对蛋白水解酶敏感的多肽抗原
OBJECTIVE: To analyze some of the biological characteristics of Mycobacterium tuberculosis polypeptide antigen (Mtb Ag) which can stimulate the proliferation of human γδT cells. Methods: The proliferation of human peripheral blood mononuclear cells (PBMCs) was stimulated with Mycobacterium tuberculosis secreted protein and Mtb Ag treated by dialysis or pronase respectively. When cultured for 10 days, the cell phenotypes were analyzed by flow cytometry. RESULTS: No significant decrease was found in the activity of γδT cells after being mobilized by Mtb Ag, while the activity of Mtb Ag treated with pronase was significantly decreased. In addition, the activity of Mtb Ag to stimulate the proliferation of γδT cells was significantly increased as compared with that of Mycobacterium tuberculosis. CONCLUSION: The component of Mtb Ag that can stimulate the proliferation of human γδT cells is a non-secreted and proteolytic enzyme-sensitive polypeptide antigen whose molecular weight is more than 100 000
