【目的】了解虫生广布拟盘多毛孢GXSTYJ03菌株侵染和杀死松突园蚧过程中酶活性变化及作用机理。【方法】通过Folin-酚法、DNS法和对硝基苯酚法分别测定含1×104、1×105、1×106、1×107和1×108个孢子/mL系列浓度的GXSTYJ03菌株孢子悬浮液产生的蛋白酶、几丁质酶和脂肪酶活力及其对松突圆蚧的致死率。【结果】GXSTYJ03菌株孢子含量越高,其产蛋白酶、几丁质酶、脂肪酶的活力也越高;蛋白酶、几丁质酶、脂肪酶的产酶高峰期和产酶活性均不相同,蛋白酶的产酶高峰期出现在发酵后第5 d、几丁质酶出现在发酵后第6 d、脂肪酶出现在发酵后第4~5d,GXSTYJ03菌株的蛋白酶活性远大于几丁质酶和脂肪酶活性;GXSTYJ03菌株5种孢子浓度产蛋白酶、几丁质酶、脂肪酶活性与对松突圆蚧毒力呈极显著正相关。【结论】1×107个孢子/mL的GXSTYJ03菌株孢子悬浮液是产酶及杀虫的最佳使用量。蛋白酶、几丁质酶、脂肪酶活性越高的菌株毒力越强,用于侵染和致死松突圆蚧的效果越好。
【Objective】 The objective of this study was to investigate the changes of enzyme activities and their mechanisms during the infection and killing of Acanthoeba imbricata in GXSTYJ03 strain. 【Method】 The spore suspension of GXSTYJ03 strain containing 1 × 10 4, 1 × 10 5, 1 × 10 6, 1 × 10 7 and 1 × 10 8 spores / mL was determined by Folin-phenol method, DNS method and p-nitrophenol method, Liquid-derived proteases, chitinase and lipase activities and their lethality to P. cingulata. 【Result】 The results showed that the higher the spore content of GXSTYJ03 was, the higher the activity of protease, chitinase and lipase was. The peak of protease, chitinase and lipase production and enzyme activity were different. The peak of enzyme production appeared on the 5th day after fermentation, the chitinase appeared on the 6th day after fermentation, and the lipase appeared on the 4th to 5th day after fermentation. The protease activity of GXSTYJ03 strain was far greater than that of chitinase and lipase Activity. The activities of protease, chitinase and lipase in the five spores of GXSTYJ03 strain were significantly and positively correlated with the virulence of Cryptomeria fortunei. 【Conclusion】 Spore suspension of GXSTYJ03 strain with 1 × 107 spores / mL is the best for enzyme production and insecticidal activity. The higher the virulence of protease, chitinase and lipase activity strains, the better the effect of infecting and killing pine scale.
