目的:建立去甲斑蝥素原位凝胶中去甲斑蝥素的含量测定方法。方法:采用HPLC法,色谱柱:Agilent ZORBAX SBC_(18)柱(150 mm×4.6 mm,5μm),流动相:乙腈-磷酸盐缓冲液(1∶9,用磷酸调p H至3.1),流速:0.8 ml·min~(-1),柱温:25℃,检测波长:210 nm,进样量:20μl。结果:去甲斑蝥素在0.02~1.00 mg·ml~(-1)浓度范围具有良好的线性关系(r=0.999 9),平均回收率为97.5%,RSD为0.98%(n=9)。结论:该方法准确、简单、重复性好,可用于去甲斑蝥素原位凝胶中去甲斑蝥素的含量测定。
Objective: To establish a method for the determination of norcantharidin in the in situ gel of norcantharidin. Methods: The HPLC method was used. The column was Agilent ZORBAX SBC 18 column (150 mm × 4.6 mm, 5 μm). The mobile phase was acetonitrile-phosphate buffer (1: 9, pH = : 0.8 ml · min ~ (-1), column temperature: 25 ℃, detection wavelength: 210 nm, injection volume: 20μl. RESULTS: Norcantharidin had a good linearity (r = 0.999 9) at a concentration range of 0.02-1.00 mg · ml -1 with an average recovery of 97.5% and a RSD of 0.98% (n = 9). Conclusion: The method is accurate, simple and reproducible. It can be used for the determination of norcantharidin in the in situ gel of norcantharidin.
