目的探讨急性游泳运动对小鼠心肌电压依赖性钾离子通道E家族成员1(KCNE1)表达水平的影响。方法雄性BALB/c小鼠42只,随机分7组,每组6只。非运动为对照组,余6组于急性游泳运动30min后即刻(A组)、1h(B组)、3h(C组)、6h(D组)、12h(E组)和24h(F组)取心肌组织;用RT-PCR和Westernblot技术分别检测各组小鼠心肌KCNE1的表达水平。结果与对照组相比,A、C和D组心肌KCNE1表达水平明显升高(P<0.05)。结论急性运动引起心肌KCNE1表达水平一过性升高,可能是交感紧张增强后心脏电生理活动发生改变的原因之一。
Objective To investigate the effect of acute swimming exercise on the expression of voltage-dependent potassium channel E family member 1 (KCNE1) in mice. Methods Forty-two male BALB / c mice were randomly divided into 7 groups with 6 mice in each group. (Group A), 1h (group B), 3h (group C), 6h (group D), 12h (group E) and 24h (group F) after swimming for 30min. Myocardial tissue was harvested. The expression of KCNE1 in each group was detected by RT-PCR and Western blot. Results Compared with the control group, the expression of KCNE1 in myocardium of A, C and D groups was significantly increased (P <0.05). Conclusion Acute exercise induced a transient increase of KCNE1 expression in myocardium, which may be one of the reasons for the change of cardiac electrophysiological activity after sympathetic tone enhancement.
