目的为了研究开发一种具有高灵敏度、高特异性的西地那非(sildenafil,SD)多克隆抗体。方法本文通过人工合成西地那非半抗原(SDH)和西地那非抗原(SDH-BSA),然后经免疫兔子,制备了SD多克隆抗体,并对制得的抗体的特异性进行了分析。结果免疫后产生了SD多克隆抗体,抗体的效价为3.8×105,线性检测范围为0.02~3.8 ng/m L,抗体除与瓦地那非具有较高的交叉反应率外,与红地那非、他地那非等结构类似物交叉反应率均较低。结论采用本方法制备的SD多克隆抗体具有较高的特异性和灵敏度,可用于SD快速检测试剂盒的开发。
Objective To study the development of a highly sensitive and highly specific polyclonal antibody against sildenafil (SD). Methods SD polyclonal antibody was prepared by artificial synthesis of SDH and SDH-BSA and then immunized with rabbit, and the specificity of the obtained antibody was analyzed . Results SD polyclonal antibody was generated after immunization. The titer of the antibody was 3.8 × 105, the linear range was 0.02-3.8 ng / m L, the antibody had higher cross-reactivity than that of vadinafil, Non-non-Hendan Fei and other structural analogues cross-reactivity rates are low. Conclusion SD polyclonal antibody prepared by this method has high specificity and sensitivity and can be used in the development of SD rapid detection kit.
