目的:以龙胆中总黄酮含量为指标,采用乙醇-硫酸铵双水相体系结合微波辅助萃取法,运用正交试验法优化出最佳的提取工艺。方法:采用紫外分光光度法,以芦丁作为对照品,测定龙胆中总黄酮的含量,利用单因素试验和正交试验设计方法优化乙醇质量分数、微波功率、萃取时间、液料比等萃取条件。结果:优化的最佳条件为:双水相的组成为35%乙醇-20%硫酸铵,微波功率420W,萃取时间25min,液料比80∶1,药材颗粒度100目。结论:本实验采用正交试验法对龙胆中的总黄酮提取工艺进行了优化,该方法合理可行,对龙胆中黄酮类化合物的提取具有一定的指导意义。
OBJECTIVE: To optimize the extraction process by the orthogonal test with ethanol-ammonium sulfate aqueous two-phase system combined with microwave-assisted extraction using the total flavonoids content of gentian as an index. Methods: The content of total flavonoids in gentian was determined by ultraviolet spectrophotometry and rutin as reference substance. The single factor experiment and orthogonal experimental design method were used to optimize the ethanol concentration, microwave power, extraction time, ratio of liquid to solid condition. Results: The optimized conditions were as follows: aqueous phase composed of 35% ethanol and 20% ammonium sulfate, microwave power of 420W, extraction time of 25 min, ratio of liquid to material of 80: 1 and particle size of 100 mesh. Conclusion: In this experiment, the orthogonal experiment was used to optimize the extraction technology of total flavonoids in Gentiana scabra. The method is reasonable and feasible, which is of guiding significance for the extraction of flavonoids from Gentiana scabra.
