用4种土壤微生物DNA提取方法提取了3种类型土壤的微生物总DNA,利用分光光度法测定4种DNA的OD260、OD280,并计算和分析DNA提取物的产率和纯度.研究发现:方法4即改良的DNA提取方法,无论是提取DNA的产率,还是提取DNA的纯度,都较其他3种方法高,说明改良的DNA提取方法能够准确有效地提取3种类型土壤的微生物DNA.
Four kinds of soil microbial DNA extraction methods were used to extract the total microbial DNA of three types of soils, and the OD260 and OD280 of four kinds of DNA were determined by spectrophotometry, and the yield and purity of DNA extracts were calculated and analyzed.The results showed that: Method 4 That is to say, the improved DNA extraction method, both for yield of DNA extraction and purity of DNA extraction, is higher than the other three methods, indicating that the improved DNA extraction method can accurately and efficiently extract microbial DNA from three types of soils.
