采用单因素和正交试验研究了氮源、蔗糖浓度和诱导子茉莉酸甲酯对蒙古黄芪愈伤组织生长和黄酮类物质积累的影响.结果表明:各因子影响黄酮积累量依次为:茉莉酸甲酯>蔗糖>水解酪蛋白>氮源;蒙古黄芪愈伤组织生长和黄酮含量积累的最佳培养基组合为:MS+NAA 1.0mg/L+6-BA 2.0mg/L+2,4-D 1.0mg/L+蔗糖50.0g/L+水解酪蛋白1.0g/L+MeJA0.02μmol/L,NH4+∶NO3-为1∶1.在这种条件下,黄酮产量达到最高为9.78mg/L.
The effects of nitrogen source, sucrose concentration and methyl jasmonate (MEK) on the callus growth and flavonoids accumulation of Astragalus membranaceus in Mongolia were studied by single factor and orthogonal experiments.The results showed that the accumulation of flavonoids in each factor were as follows: jasmonic acid Methyl ester> sucrose> hydrolyzed casein> nitrogen source; the best medium for the accumulation of callus and the accumulation of flavonoids in Astragalus mongolia was MS + NAA 1.0 mg / L + 6-BA 2.0 mg / D 1.0mg / L + sucrose 50.0g / L + hydrolyzed casein 1.0g / L + MeJA 0.02μmol / L, NH4 +: NO3- 1: 1. Under these conditions, the highest yield of flavonoids was 9.78mg / L.
