目的:对纤维素酶用于黄芪有效成分提取的酶解参数进行优选。方法:通过正交实验,以黄芪甲苷、总浸出物为指标,采用HPLC等方法测定指标成分的含量,综合评价酶解工艺的优劣。结果:最佳酶解条件为:纤维素酶用量0.1%、pH值6、酶解温度50℃、酶解时间90m in。结论:本方法简便有效,黄芪有效成分提取率高,为对黄芪深入研究奠定基础。
OBJECTIVE: To optimize the enzymatic hydrolysis parameters of cellulase for the extraction of active ingredients of Astragalus membranaceus. Methods: Through orthogonal experiment, using astragaloside and total extract as indexes, the contents of index components were determined by HPLC and so on, and the advantages and disadvantages of enzymolysis process were comprehensively evaluated. Results: The optimal conditions for enzymolysis were as follows: cellulase dosage 0.1%, pH value 6, enzymolysis temperature 50 ℃, enzymolysis time 90m in. Conclusion: The method is simple and effective, the extraction rate of active ingredients of Astragalus is high, which lays the foundation for further study of Astragalus.
