目的:观察加味参附汤对肾阳虚肺纤维化大鼠模型的病变程度及caspase-3和Bcl-2在模型大鼠肺内表达的影响。方法:大鼠60只随机分为正常组、模型组和干预组。模型组和干预组制作肾阳虚肺纤维化大鼠模型,造模成功后干预组给予加味参附汤治疗,常规石蜡切片,HE染色,观察肺泡炎及肺纤维化程度,免疫组织化学技术检测caspase-3和Bcl-2蛋白在各组大鼠肺组织中的表达。结果:caspase-3和Bcl-2主要在肺泡上皮细胞、肺泡巨噬细胞及浸润的炎症细胞中表达,第28天,模型组、干预组caspase-3和Bcl-2蛋白表达增强,均高于正常组(P<0.05)。第42天,干预组大鼠细胞caspase-3表达较模型组降低(P<0.05),而Bcl-2表达在干预组中增强,高于模型组(P<0.05)。结论:在大鼠肾阳虚肺纤维化的发展过程中,肺组织细胞凋亡起重要作用,加味参附汤可用通过调控caspase-3和Bcl-2蛋白的表达而减少细胞凋亡,从而减缓肾阳虚肺纤维化的进展。
OBJECTIVE: To observe the effect of Jiawei Shenfu Decoction on the pathological changes and the expression of caspase-3 and Bcl-2 in the lungs of rats with kidney-yang deficiency and pulmonary fibrosis. Methods: Sixty rats were randomly divided into normal group, model group and intervention group. The model group and the intervention group were made into the rat model of kidney-yang deficiency pulmonary fibrosis. After the model was established, the intervention group was treated with Jiawei Shenfu decoction. The paraffin sections and HE staining were used to observe the degree of alveolitis and pulmonary fibrosis. Immunohistochemistry Expression of caspase-3 and Bcl-2 in lung tissue of rats in each group. Results: The expressions of caspase-3 and Bcl-2 were mainly in alveolar epithelial cells, alveolar macrophages and infiltrating inflammatory cells. On the 28th day, the expressions of caspase-3 and Bcl-2 protein increased Normal group (P <0.05). On the 42nd day, the expression of caspase-3 in the intervention group was lower than that in the model group (P <0.05), while the expression of Bcl-2 in the intervention group was higher than that in the model group (P <0.05). CONCLUSION: Apoptosis of lung tissue plays an important role in the development of pulmonary fibrosis in rats with deficiency of kidney-yang. Jiawei Shenfu decoction can reduce apoptosis by regulating the expression of caspase-3 and Bcl-2 protein, Progress in Kidney Yang Deficiency Pulmonary Fibrosis.
