为了研究不同品种甘薯在潮霉素以及basta选择下的耐受性,为甘薯的遗传转化提供依据,本试验选取了四川省常用甘薯栽培品种(材料),川薯164与西成薯007,以叶片为外植体,分别接种于含有6-BA、NAA及不同浓度梯度的潮霉素及basta的培养基上,统计分析得出不同基因型甘薯的最适抗性压。结果表明,川薯164与西成薯007叶片抗hyg的最适选择压分别为20~30,15~20 mg/L,川薯164与西成薯007叶片抗basta的最适选择压均为1.0 mg/L。
In order to study the tolerance of different varieties of sweet potato under hygromycin and basta selection, this study provided the basis for the genetic transformation of sweet potato. In this experiment, we selected Sichuan sweet potato cultivars (materials), Sichuan potato 164 and Xi Cheng potato 007, The explants were inoculated on the medium of hygromycin and basta with 6-BA, NAA and different concentration gradients, respectively. The optimum pressure of different genotype sweet potato was obtained by statistical analysis. The results showed that the optimum selection pressure of resistance to hyg between Chuanshu 164 and Xi Cheng potato 007 was 20-30 and 15-20 mg / L, respectively. The optimum selection pressure of anti-basta of Chuanshu 164 and Xi Cheng potato 007 was 1.0 mg / L.
