目的:探讨青蒿琥酯对肝细胞保护作用的机理。方法:采用四氯化碳致小鼠肝损伤模型,利用电镜细胞化学技术和体视学技术研究钙离子在肝细胞的分布情况。结果:模型组钙离子在胞浆中的数密度较正常组和青蒿琥酯高剂量组均有明显增高,差异有非常显著性意义(P<0.01);模型组与青蒿琥酯低剂量组小鼠胞浆钙离子数密度比较,差异无显著性意义(P>0.05);青蒿琥酯高剂量组胞浆钙离子数密度与秋水仙素组比较,差异有显著性意义(P<0.05)。结论:青蒿琥酯具有稳定细胞膜、内质网和线粒体等膜性结构的作用,使影响细胞生理活性的钙离子在胞内外处于稳态的水平,从而阻挡了因钙离子的失衡而启动的脂质过氧化反应对肝细胞的损伤。
Objective: To investigate the mechanism of artesunate on liver cell protection. Methods: CCl4-induced liver injury model was used in mice to study the distribution of calcium in hepatocytes by electron microscopy and stereological techniques. Results: The number density of calcium ion in cytoplasm in model group was significantly higher than that in normal group and artesunate high dose group (P <0.01). The model group and artesunate low dose There was no significant difference in the number density of cytoplasmic calcium between the two groups (P> 0.05). The number density of cytoplasmic calcium in high dose artesunate group was significantly higher than that of colchicine group (P < 0.05). CONCLUSIONS: Artesunate stabilizes the membranous structure such as cell membrane, endoplasmic reticulum and mitochondria so that the calcium ions that affect the physiological activity of cells are at a steady state level both inside and outside the cell, thus blocking the initiation of calcium ion imbalance Lipid peroxidation of liver cell damage.
