目的探讨转录调控因子Twist蛋白、血管内皮生长因子(VEGF)、上皮型钙黏附蛋白(E-cad)在乳腺癌中的表达及其相关性。方法采用SP免疫组织化学法检测70例乳腺癌组织(35例癌旁组织作为对照)中Twist、VEGF、E-cad的表达,并对其临床病理特点进行分析。结果 Twist、VEGF在乳腺癌组织中的表达阳性率(72.9%、75.8%)高于癌旁组织(6.0%、8.5%),差异有统计学意义(P<0.01),E-cad在乳腺癌组织中的表达阳性率(15.7%)低于癌旁组织(71.4%),差异有统计学意义(P<0.05);Twist蛋白、VEGF阳性表达率与组织病理学分级、TNM分期、腋窝淋巴结转移有相关性(P<0.05),与患者的年龄、肿瘤直径无相关性(P<0.05);E-cad阴性表达率与患者的组织病理分级、TNM分期、腋窝淋巴结转移有相关性(P<0.05),与患者的年龄、首次肿瘤大小无相关性(P<0.05);Twist与VEGF阳性表达呈正相关,与E-cad表达呈负相关。结论 Twist和VEGF在乳腺癌组织中呈高表达,而E-cad呈低表达。Twist可能通过上调VEGF的表达,同时下调E-cad的表达,从而促进乳腺癌的浸润、转移,并在其中起重要作用。
Objective To investigate the expression of Twist protein, vascular endothelial growth factor (VEGF) and E-cadherin in breast cancer and its correlation. Methods SP immunohistochemistry was used to detect the expression of Twist, VEGF and E-cad in 70 cases of breast cancer (35 cases of paracancerous tissues as control), and the clinicopathological features were analyzed. Results The positive expression rates of Twist and VEGF in breast cancer tissues were significantly higher than those in adjacent tissues (72.9%, 75.8% vs 6.0%, 8.5%, P <0.01) The positive expression rate of Twist protein, VEGF and histopathological grade, TNM staging, axillary lymph node metastasis (P <0.05), the positive rate of expression in tissue (15.7%) was lower than that in paracancerous tissue There was no correlation between the expression of E-cad and the histopathological grade, TNM stage and axillary lymph node metastasis (P <0.05), but not with age and tumor diameter (P <0.05) 0.05). There was no correlation between age and tumor size (P <0.05). Twist was positively correlated with VEGF expression and negatively correlated with E-cadherin expression. Conclusion Twist and VEGF are highly expressed in breast cancer tissues, while E-cad is low expression. Twist may play an important role in the invasion and metastasis of breast cancer by up-regulating the expression of VEGF and down-regulating the expression of E-cadherin.
