目的 探讨鼻咽癌细胞在乏氧 -再氧合过程中乏氧诱导因子 1α(HIF 1α)和血管内皮生长因子 (VEGF)基因mRNA水平表达的变化。方法 鼻咽鳞癌细胞系 (CNE)分别在富氧、乏氧 8h、乏氧 16h和乏氧 16h后再氧合 4h 4个不同条件下培养后 ,应用反转录聚合酶链反应 (RT PCR)方法半定量测定HIF 1α和VEGF基因的mRNA水平的表达。结果 在 4个不同培养条件下的CNE细胞的HIF 1α基因的mRNA水平表达无明显变化 ,而VEGF基因的mRNA水平表达经乏氧 8h和乏氧 16h培养后均明显升高 ,在乏氧 16h后再氧合 4h培养后的表达则显著下降。结论 乏氧能引起鼻咽癌细胞的VEGF基因mRNA水平高表达 ,而对HIF 1α基因的mRNA表达水平影响不明显 ,提示乏氧可能不是通过HIF 1α基因的转录途径来诱发VEGF基因的高表达而达到提高肿瘤侵袭性的目的。
Objective To investigate the changes of mRNA expression of hypoxia inducible factor 1α (HIF 1α) and vascular endothelial growth factor (VEGF) in nasopharyngeal carcinoma cells during hypoxia - reoxygenation. Methods Nasopharyngeal squamous cell carcinoma (CNE) cells were cultured in four different conditions: oxygen enrichment, hypoxia for 8 h, hypoxia for 16 h and reoxygenation for 4 h after hypoxia, respectively. Reverse transcriptase polymerase chain reaction ) Method was used to semi-quantitatively determine the mRNA expression of HIF-1α and VEGF genes. Results There was no significant change in mRNA expression of HIF 1α gene in CNE cells under 4 different culture conditions. However, the mRNA expression of VEGF gene was significantly increased after 8 h of hypoxia and 16 h of hypoxia. After 16 h of hypoxia Oxygenation after 4h cultured significantly decreased. CONCLUSION: Hypoxia can induce high expression of VEGF mRNA in NPC cells, but not HIF-1α mRNA expression, suggesting that hypoxia may not induce the high expression of VEGF gene through the transcriptional pathway of HIF-1α gene To achieve the purpose of improving tumor aggressiveness.
