目的:分离制备毛冬青三萜皂苷ilexsaponin A1(IPTS-1),并探讨其药理活性,为阐明毛冬青的活性成分提供实验依据。方法:综合运用色谱法和重结晶法等分离纯化方法进行化合物的制备,采用薄层对照及核磁波谱分析的方法对化合物的结构进行鉴定。通过凝血时间、出血时间、小鼠耳廓微循环实验和大鼠FeCl3诱导血栓实验对IPTS-1的抗凝、活血、抗血栓作用进行评估和探讨。结果:与对照组比较,IPTS-1各剂量均能显著延长小鼠出血时间和凝血时间(P<0.05),且作用呈剂量相关性。IPTS-1可加速小鼠耳廓微循环血流速度,减少FeCl3诱导的大鼠腹主动脉血栓生成质量。结论:毛冬青三萜皂苷IPTS-1具备一定的抗凝、活血、抗血栓作用,可能为毛冬青心血管药理作用的物质基础之一。
OBJECTIVE: To isolate and prepare ilexsaponin A1 (IPTS-1), and to explore the pharmacological activity of triterpene saponins. Methods: The compounds were prepared by separation and purification methods such as chromatography and recrystallization. The structures of the compounds were identified by TLC and NMR spectroscopy. The anticoagulant, blood-activating and antithrombotic effects of IPTS-1 were evaluated and evaluated by clotting time, bleeding time, mouse auricle microcirculation and rat FeCl3-induced thrombosis. Results: Compared with the control group, all doses of IPTS-1 significantly prolonged the bleeding time and clotting time (P <0.05), and the effect was dose-dependent. IPTS-1 can accelerate the auricle microcirculation blood flow velocity and reduce the FeCl3-induced abdominal aorta thrombosis in rats. Conclusion: Triterpenoid saponin IPTS-1 possesses certain anticoagulation, blood-activating and antithrombotic effects and may be one of the material bases for the cardiovascular pharmacological effects of Mao Dongqing.
