探索检测饲料、蛋黄和鸡肝中长链饱和、单烯及多烯键脂肪酸的前处理步骤,样品匀浆后,用氯仿-丙酮混合溶剂提取,以氢氧化钾-甲醇酯化后使生成相应的脂肪酸甲酯,再转溶于异辛烷中,采用不分流进样技术注入气相色谱仪中,经毛细管柱分离,用火焰离子化检测器检测。应用该法能检测出含量较低、具有生理活性意义但易于氧化的ω-3长链多烯键脂肪酸,如二十碳五烯酸(EPA)、二十二碳五烯酸(DPA)、二十二碳六烯酸(DHA)。
To explore the detection of feed, egg yolk and chicken liver long-chain saturated, monoene and polyene fatty acid pretreatment steps, the sample was homogenized, with chloroform - acetone mixed solvent extraction, potassium hydroxide - methanol esterification to generate the corresponding Fatty acid methyl ester, and then dissolved in isooctane, using splitless injection technology into the gas chromatograph, the capillary column separation, with flame ionization detector detection. This method can be used to detect low, physiologically active but readily oxidizable omega-3 long chain polyunsaturated fatty acids, such as EPA, docosapentaenoic acid (DPA) Docosahexaenoic acid (DHA).
