考查培养基中Co、I元素的含量及培养基初始pH对小麦胚芽愈伤组织VE积累的影响,建立VE积累和细胞生长的动力学模型。以次生代谢产物VE积累最多为目标,通过试验优化确定Co、I的含量和初始pH值的最佳组合。结果表明,当N6培养基中CoCl20.025mg/L、KI0.80mg/L、pH5.8时,有利于VE积累。VE积累的动力学模型的拟合度为98.11%,细胞增重的动力学模型拟合度为99.40%,非线性关系良好。有益元素Co和I对小麦胚芽愈伤组织VE积累有影响,I元素较Co元素的影响显著。
The contents of Co and I in the medium and the initial pH of medium were used to investigate the effect of the accumulation of VE on the accumulation of VE and the kinetic model of cell growth. With the goal of maximizing the accumulation of secondary metabolites VE, the optimal combination of Co, I content and initial pH was determined experimentally. The results showed that when NCl medium CoCl20.025mg / L, KI0.80mg / L, pH5.8, is conducive to the accumulation of VE. The fitted kinetic model of VE was 98.11%, and the kinetic model of cell weight gain was 99.40%. The nonlinearity was good. The beneficial elements, Co and I, affected the accumulation of VE in wheat embryo callus, and the effect of I element was more significant than that of Co element.
