目的探讨组蛋白去乙酰化酶1(HDAC1)、转录因子E2F1在人肾透明细胞癌中的表达和意义。方法应用免疫组织化学法检测51例肾透明细胞癌组织、24例癌旁正常组织中HDAC1、E2F1的蛋白表达情况。RT-PCR方法检测31例肾透明细胞癌组织、14例癌旁正常组织中HDAC1、E2F1的mRNA相对表达情况。观察HDAC1及E2F1的表达与临床病理特征的关系。结果免疫组化结果显示:HDAC1蛋白在肾透明细胞癌组织中的阳性表达率,高于癌旁正常组织(P<0.05);E2F1蛋白在肾透明细胞癌组织中的阳性表达率,高于癌旁正常组织(P<0.05);HDAC1与E2F1的蛋白表达与患者性别、年龄及瘤体大小无关(P>0.05),而与组织学分级及临床分期有关(P<0.05)。RT-PCR结果显示:HDAC1mRNA在肾透明细胞癌组织中的相对表达量,高于癌旁正常组织(P<0.05);E2F1mRNA在肾透明细胞癌组织中的相对表达量,高于癌旁正常组织(P<0.05);HDAC1与E2F1的mRNA相对表达量与患者性别、年龄及瘤体大小无关(P>0.05),而与组织学分级及临床分期有关(P<0.05)。结论 HDAC1与E2F1在肾透明细胞癌中的表达高于癌旁正常组织,且与组织学分级及临床分期有关,并随组织恶性程度的增加其表达水平逐渐增加,提示与肾透明细胞癌的进展有密切关系。
Objective To investigate the expression and significance of histone deacetylase 1 (HDAC1) and transcription factor E2F1 in human renal clear cell carcinoma. Methods The expression of HDAC1 and E2F1 protein in 51 cases of clear cell renal cell carcinoma and 24 cases of adjacent normal tissues were detected by immunohistochemistry. RT-PCR was used to detect the mRNA expression of HDAC1 and E2F1 in 31 cases of renal clear cell carcinoma tissues and 14 cases of adjacent normal tissues. To observe the relationship between the expression of HDAC1 and E2F1 and clinicopathological features. Results The positive rate of HDAC1 protein expression in renal clear cell carcinoma tissue was higher than that in adjacent normal tissue (P <0.05). The positive expression rate of E2F1 protein in renal clear cell carcinoma tissue was higher than that in carcinoma (P <0.05). The protein expression of HDAC1 and E2F1 was not related to the sex, age and size of tumor (P> 0.05), but correlated with histological grade and clinical stage (P <0.05). The results of RT-PCR showed that the relative expression of HDAC1 mRNA in renal clear cell carcinoma tissues was higher than that in adjacent normal tissues (P <0.05), and the relative expression of E2F1 mRNA in renal clear cell carcinoma tissues was higher than that in adjacent normal tissues (P <0.05). The relative expression of HDAC1 and E2F1 mRNA was not related to the gender, age and tumor size (P> 0.05), but correlated with histological grade and clinical stage (P <0.05). Conclusions The expression of HDAC1 and E2F1 in renal clear cell carcinoma is higher than that in adjacent normal tissues. The expression of HDAC1 and E2F1 is related to the histological grade and clinical stage. The expression of HDAC1 and E2F1 gradually increases with the progression of renal clear cell carcinoma There is a close relationship.
