目的 比较粘着斑激酶 (FAK)和整合蛋白 (integrin)α5、β1亚基在胃癌及癌旁组织中的表达与肿瘤细胞分化、浸润、转移等生物学行为的关系及相互关系。方法 用免疫组织化学亲合素生物素法 (ABC)检测 75例胃癌及癌旁组织石蜡标本FAK和整合蛋白α5、β1亚基蛋白的表达水平。结果 FAK、整合蛋白α5亚基和整合蛋白 β1亚基的表达为 或 分别是胃癌组织 5 7%、2 9%和 2 5 % ,癌旁组织 4%、76%和 84% ;低分化胃癌组织 68%、18%和 16% ,高中分化组织 42 %、45 %和 39% ;有淋巴结转移的组织 77%、31%和 31% ,无转移的组织 36%、2 8%和 19% ;浸润程度较深的组织 68%、2 9%和 2 5 % ,浸润程度较浅的组织 19%、31%和 2 5 %。结论 FAK较整合蛋白α5、β1亚基可以更有效地衡量胃癌的发生与发展 ,FAK的表达量可作为胃癌发生、发展过程中一种较有价值的病理指标。FAK可能既是一种转化相关酶又是一种演变相关酶。
Objective To compare the relationship between the expression of focal adhesion kinase (FAK) and integrin α5 and β1 subunits in gastric cancer and adjacent tissues and their biological behavior, such as tumor cell differentiation, invasion, and metastasis. Methods The expression levels of FAK and integrin α5 and β1 subunits in paraffin-embedded tissues were measured by immunohistochemical avidin biotin method (ABC). Results The expressions of FAK, integrin α5 subunit and integrin β1 subunit were or were respectively 57%, 29% and 25% in gastric cancer tissues, 4%, 76% and 84% in adjacent tissues; poorly differentiated gastric cancer tissues. 68%, 18%, and 16%, 42%, 45%, and 39% of high-grade differentiated tissues, 77%, 31%, and 31% of tissues with lymph node metastases, 36%, 28%, and 19% of tissues without metastasis; The deeper tissues were 68%, 29%, and 25%, and the infiltrating tissues were 19%, 31%, and 25%. Conclusion FAK is more effective than integrin α5 and β1 subunits in measuring the occurrence and development of gastric cancer. The expression of FAK can be used as a valuable pathological indicator in the development and progression of gastric cancer. FAK may be both a transformation-related enzyme and an evolution-related enzyme.
