目的探讨腹腔镜CO2气腹对人宫颈癌CaSki细胞体外增殖能力的影响。方法建立腹腔镜CO2气腹体外模型,以不同压力(0、7、14 mmHg)的CO2气腹对人宫颈癌CaSki细胞分别处理不同时间(1、2、3、4 h),设未处理组为对照。采用流式细胞仪检测各组细胞周期分布及增殖指数的变化,采用免疫细胞化学法分别检测处理后各组细胞PCNA和Ki-67的表达情况。结果①7 mmHg CO2气腹主要增加G2~M期细胞比例,而14 mmHg CO2气腹主要增加S期细胞比例。②CaSki细胞的增殖指数与一定的压力和时间成正相关。③CO2气腹在一定的压力和作用时间范围能上调PCNA和Ki-67在CaSki细胞中的表达。结论CO2气腹可提高人宫颈癌CaSki细胞体外增殖能力,从而实现其促肿瘤细胞生长的生物学效应。
Objective To investigate the effect of laparoscopic CO2 pneumoperitoneum on proliferation of human cervical carcinoma CaSki cells in vitro. Methods The laparoscopic CO2 pneumoperitoneum model was established. The CaSki cells were treated with CO2 pneumoperitoneum at different pressures (0, 7, 14 mmHg) for different times (1,2,3,4 h) As a control. The changes of cell cycle distribution and proliferation index in each group were detected by flow cytometry. The expression of PCNA and Ki-67 in each group were detected by immunocytochemistry. Results ① 7 mmHg CO2 pneumoperitoneum increased the proportion of G2 to M phase cells, while 14 mmHg CO2 pneumoperitoneum increased the proportion of S phase cells. ②CaSki cell proliferation index and a certain pressure and time is positively correlated. ③CO2 pneumoperitoneum can up-regulate the expression of PCNA and Ki-67 in CaSki cells under certain pressure and action time range. Conclusion CO2 pneumoperitoneum can increase the proliferation of human cervical cancer CaSki cells in vitro and thus achieve the biological effect of promoting the growth of tumor cells.
