目的探讨未发生梗阻性肺动脉高压的左向右分流先天性心脏病儿童骨骼肌血管是否有AT2受体表达,及肺循环AngⅡ受体的表达。方法20例左向右分流先心病患儿术中取肺、骨骼肌活检。用RT-PCR和免疫组化的方法检测这些骨骼肌中的AT2受体;并对肺组织中AT1和AT2受体的mRNA作半定量分析。结果RT-PCR在这些骨骼肌中均检出AT2受体的mRNA;这些骨骼肌血管的AT2受体免疫组化染色均为阳性。这些患儿肺组织中AT2受体的mRNA水平比AT1受体高(P<0.05)。结论左向右分流先心病儿童骨骼肌血管有AT2受体表达;肺循环AT2受体表达水平比AT1受体高。
Objective To investigate the expression of AT2 receptor in skeletal muscle of children with left-to-right shunt congenital heart disease without obstructive pulmonary hypertension and the expression of Ang Ⅱ receptor in pulmonary circulation. Methods Twenty patients with left-to-right shunt with congenital heart disease underwent lung biopsy and skeletal muscle biopsy. The AT2 receptors in these skeletal muscles were detected by RT-PCR and immunohistochemistry. The mRNAs of AT1 and AT2 receptors in lung tissues were semi-quantitatively analyzed. Results RT-PCR detected AT2 receptor mRNA in these skeletal muscles; all these skeletal muscle vessels showed positive AT2 receptor immunohistochemical staining. AT2 receptor mRNA levels were higher in lung tissue than in AT1 recipients (P <0.05). Conclusion The expression of AT2 receptor in skeletal muscle of left-right shunt children with congenital heart disease was higher than that of AT1 receptor.
