目的:研究对苯二酚在碱性条件下产生半醌自由基及其对耐甲氧西林的金黄色葡萄球菌(MRSA)、甲氧西林敏感葡萄球菌(MSSA)、BL21大肠杆菌3种菌的抑菌活性。方法:应用电子自旋共振(ESR)检测半醌昆自由基;应用高效液相色谱法(HPLC)检测对苯二酚在碱性条件下的反应产物;应用打孔法,研究对苯二酚在碱性条件下对MRSA、MSSA、BL21大肠杆菌3种菌的抑菌作用,并采用了96孔板法研究其对这3种菌的最小抑菌浓度(MIC);同时通过测量这3细菌的生长曲线来比较对苯二酚在pH=7.5与pH=6条件下的抑菌活性。结果:半醌自由基抑菌作用较对苯二酚和对苯二酚在碱性条件下产生的最终产物都强。对苯二酚碱溶液对MRSA、MSSA、BL21大肠杆菌的MIC值分别为0.081、0.081、0.649mg/mL,而对苯二酚水溶液对MRSA、MSSA、BL21的MIC值分别为0.325、0.325、1.298mg/mL。结论:对苯二酚在碱性条件下的抑菌作用远远强于其在中性或者偏酸性条件下。
OBJECTIVE: To study the effect of hydroquinone on the production of semiquinone free radicals and their resistance to methicillin-resistant Staphylococcus aureus (MRSA), methicillin-resistant staphylococcus (MSSA) and BL21 E. coli under alkaline conditions Antibacterial activity. Methods: Electron spin resonance (ESR) was used to detect the free radicals of quinonequinone. The reaction products of hydroquinone in alkaline solution were detected by high performance liquid chromatography (HPLC). The effects of hydroquinone The antibacterial activities of the three kinds of bacteria against MRSA, MSSA and BL21 were tested under alkaline conditions. The minimum inhibitory concentration (MIC) of the three kinds of bacteria was studied by 96-well plate method. The growth curves were compared for the antimicrobial activity of hydroquinone at pH = 7.5 and pH = 6. Results: The antibacterial activity of semiquinone radicals was stronger than that of hydroquinone and hydroquinone in alkaline conditions. The MIC values of hydroquinone solution against MRSA, MSSA and BL21 were 0.081, 0.081 and 0.649 mg / mL, respectively. The MIC values of hydroquinone solution against MRSA, MSSA and BL21 were 0.325, 0.325 and 1.298, respectively mg / mL. Conclusion: The bacteriostatic effect of hydroquinone in alkaline conditions is much stronger than that in the neutral or acidic conditions.
