目的:建立HPLC-ELSD方法测定盐酸大观霉素的含量。方法:以Grace-Apollo C_(18)(150 mm×4.6 mm,5μm)为色谱柱,50 mmol·L~(-1)乙酸铵溶液(pH7.5)-甲醇(92:8)为流动相,流速为0.6 ml·min~(-1),雾化室温度:30℃,漂移管温度:70℃。雾化气体(空气)压力:0.4 Mpa。结果:盐酸大观霉素在0.101~0.504 mg·ml~(-1)范围内,呈良好的线性关系(r=0.999 0),平均回收率为98.5%(RSD=1.6%,n=9)。结论:该方法操作简便、快速,结果准确可靠,可应用于盐酸大观霉素的含量测定。
Objective: To establish HPLC-ELSD method for the determination of spectinomycin hydrochloride. METHODS: The mobile phase consisted of 50 mmol·L -1 ammonium acetate solution (pH 7.5) -methanol (92: 8) with a column of Grace-Apollo C 18 (150 mm × 4.6 mm, 5 μm) , Flow rate of 0.6 ml · min -1, atomization chamber temperature: 30 ℃, drift tube temperature: 70 ℃. Atomizing air (air) pressure: 0.4 Mpa. Results: Spectinomycin hydrochloride showed a good linear relationship (r = 0.999 0) with an average recovery of 98.5% (RSD = 1.6%, n = 9) in the range of 0.101-0.504 mg · ml -1. Conclusion: The method is simple, rapid and accurate. It can be applied to the determination of spectinomycin hydrochloride.
