目的 研究二甲基奶油黄诱发的大鼠肝癌的组织学改变。方法 设对照组 (饲料中无DAB)和实验组 (饲料中含DAB) ,4周至 16周间分批处死动物 ,光镜和电镜下观察肝脏病变情况。结果 整个过程分为非特异性炎症、肝硬化、肝癌 3个阶段。非特异性炎症阶段光镜下改变为肝细胞气球样变性、炎性细胞浸润。肝硬化阶段表现为肝细胞和胆管腺瘤样增生 ,假小叶形成 ,肝组织炎性细胞浸润。肝癌有肝细胞癌、胆管细胞癌和混合细胞癌 3种组织类型。超微结构显示癌细胞间连接复合体减少 ,核膜增厚 ,线粒体减少。胆管细胞癌中发现致密核心颗粒。结论 在由二甲基奶油黄诱发的大鼠肝肿瘤中 ,可发现组织学改变及致密核心颗粒
Objective To study the histological changes of hepatocarcinoma induced by dimethyl-cream yellow in rats. Methods The control group (no DAB in feed) and experimental group (DAB in feed) were sacrificed in batches in 4 weeks to 16 weeks. The pathological changes of liver were observed under light and electron microscope. Results The whole process was divided into non-specific inflammation, cirrhosis, liver cancer in three stages. Non-specific inflammation stage changed to hepatocyte balloon degeneration and inflammatory cell infiltration by light microscope. Liver cirrhosis stage showed hepatocellular and bile duct adenoma-like hyperplasia, pseudolobule formation, liver inflammatory cell infiltration. Liver cancer has hepatocellular carcinoma, cholangiocarcinoma and mixed cell carcinoma three kinds of tissue types. The ultrastructure showed a decrease in the junctional complex of cancer cells, a thickening of the nuclear membrane and a decrease of mitochondria. Dense core particles are found in cholangiocarcinoma. Conclusions In rat liver tumors induced by dimethyl-cream-yellow, histological changes and dense core particles
