目的 探讨人粘连蛋白 (FN)基因在胰腺癌发生和转移中的作用。方法 应用博道40 96型cDNA基因芯片和原位杂交技术检测 18例胰腺癌标本、8例慢性胰腺炎组织FN基因表达。结果 FN基因在 18例胰腺癌基因芯片检测中表达显著增高 (Cy5 /Cy3 >2 .0 ,P <0 .0 1) ,伴有区域淋巴结转移时增高更加明显 ;在慢性胰腺炎中的表达与正常组织比较差异无显著性 (Cy5 /Cy3 <2 .0 ,P >0 .0 5 ) ,与原位杂交结果相同。结论 基因芯片技术能够为胰腺癌的相关基因分析提供特异和可靠的数据 ;人胰腺组织中FN基因表达升高可能与胰腺癌的发生和转移有关。
Objective To investigate the role of human fibronectin (FN) in the genesis and metastasis of pancreatic cancer. Methods The mRNA expression of FN in 18 cases of pancreatic cancer and 8 cases of chronic pancreatitis was detected by using cDNA microarray 40 96 and in situ hybridization. Results The expression of FN gene was significantly increased in 18 cases of pancreatic cancer gene chip detection (Cy5 / Cy3> 2.0, P <0.01), with more obvious increase of regional lymph node metastasis; in chronic pancreatitis, There was no significant difference in normal tissues (Cy5 / Cy3 <2.0, P> 0.05), which was the same as in situ hybridization. Conclusion Gene chip technology can provide specific and reliable data for the related gene analysis of pancreatic cancer. The increase of FN gene expression in human pancreatic tissue may be related to the occurrence and metastasis of pancreatic cancer.
