目的:探讨不同剂量黄芪对脾虚发热大鼠模型的作用。方法:大鼠随机分为正常对照组、脾虚发热组、黄芪大、中、小剂量组共5组。采用饮食失节+游泳疲劳制备脾虚模型,实验第18天开始,黄芪各组大鼠分别按剂量2.16g/kg(大)、1.08g/kg(中)、0.54g/kg(小)给予灌胃,正常组和脾虚发热组给予同体积蒸馏水,每日1次,连续5天。第22d的AM 8:00,各实验组大鼠ipLPS(80μg/kg),正常对照组ip等量生理盐水,观测每周各组大鼠的外观行为、LPS注射后不同时间点的体温和血IL-1β、IL-2、GAS、CCK、B-EP、SS。结果:与正常组相比,脾虚发热组大鼠在ip LPS后60、120、180min时的体温出现明显升高(P<0.05),外观行为积分值显著升高,血中SS、IL-1β、IL-2含量见显著升高(P<0.05)。与脾虚发热组比较,黄芪大剂量组外观行为的部分指标有所改善;黄芪三个剂量组60min后的体温均呈现不同程度的降低,其中大、中剂量组的多点体温呈显著降低(P<0.05);大剂量组IL-2和GAS含量均见显著降低(P<0.05),中剂量组SS/CCK含量呈显著降低/升高(P<0.05),小剂量组GAS和SS含量均见显著性降低(P<0.05)。较之于中、小剂量组,黄芪大剂量组IL-2含量降低明显(P<0.05)。结论:黄芪对脾虚发热大鼠模型有不同程度的调节作用,综合效应指标则以黄芪大剂量作用明显。
Objective: To investigate the effects of Astragalus membranaceus in different doses on rat model of spleen-deficiency fever. Methods: The rats were randomly divided into normal control group, spleen deficiency fever group, Astragalus large, medium and low dose group of 5 groups. The model of spleen deficiency was established by dietary nocturnal and swimming fatigue, and the rats in each group were given intragastric administration of 2.16g / kg (big), 1.08g / kg (middle) and 0.54g / kg , Normal group and spleen deficiency group were given the same volume of distilled water once a day for 5 consecutive days. At 8:00 AM on the 22nd day, ipLPS (80μg / kg) in the experimental group and normal saline in the normal control group were observed. The appearance and behavior of rats in each group were observed. Body temperature and blood at different time points after LPS injection IL-1β, IL-2, GAS, CCK, B-EP, SS. Results: Compared with the normal group, the body temperature of spleen-heat-induced rats increased significantly (P <0.05) at 60, 120 and 180 minutes after ip LPS, and the scores of appearance behavior increased significantly. The levels of SS, IL-1β , IL-2 content was significantly increased (P <0.05). Compared with the spleen-febrile group, some indicators of the external appearance behavior of the high-dose Astragalus group were improved. The body temperature of the Astragalus-Astragalus-Astragalus-Astragalus group after 60min decreased to some extent, (P <0.05). The levels of IL-2 and GAS in high-dose group were significantly decreased (P <0.05), and the content of SS / CCK in medium dose group was significantly decreased / See significant reduction (P <0.05). Compared with medium and low dose group, Astragalus high dose group IL-2 content decreased significantly (P <0.05). Conclusion: Astragalus can modulate the rat model of spleen-deficiency fever to some extent, and the comprehensive effect index is obviously treated with high dose of astragalus.
