目的探讨胡椒碱(PIP)对胰岛素抵抗综合征(IRS)大鼠甘油三酯(TG)、游离脂肪酸(FFA)、肿瘤坏死因子(TNF-α)、瘦素(Leptin)的作用。方法采用高脂高糖高盐饲料喂养制备IRS大鼠模型。每4周观察各组空腹TG,第4周开始给药,连续给药8周后,分别采用GPO-PAP法、比色法检测TG和FFA水平,ELISA法检测TNF-α、Leptin水平。结果给药8周后,模型大鼠血清TG、FFA、TNF-α、Leptin水平明显升高,不同剂量PIP使TG、FFA、TNF-α、Leptin水平下降。结论 PIP明显改善IRS模型大鼠的脂代谢异常情况,能降低IRS模型大鼠TG和FFA水平,其作用机制可能与下调细胞因子TNF-α、Leptin有关。
Objective To investigate the effects of piperine on triglyceride (TG), free fatty acid (FFA), tumor necrosis factor (TNF-α) and leptin in rats with insulin resistance syndrome (IRS). Methods IRS rats were fed with high-fat, high-sugar and high-salt diet. The fasting TG was observed every 4 weeks, and began to be administered from the 4th week. After continuous administration for 8 weeks, the levels of TG and FFA were detected by GPO-PAP method and the levels of TNF-α and Leptin by ELISA. Results After 8 weeks of administration, the levels of TG, FFA, TNF-α and Leptin in the model rats were significantly increased. The levels of TG, FFA, TNF-α and Leptin were decreased with different doses of PIP. Conclusion PIP can significantly improve the abnormal lipid metabolism in IRS model rats and decrease the levels of TG and FFA in IRS rats. The mechanism may be related to the down-regulation of cytokines TNF-α and Leptin.
