目的:研究烟酰胺磷酸核糖转移酶(Nicotinamide phosphoribosyltransferase,NAMPT)在胶质瘤中的表达和临床意义,并进行GO和Pathway注释.方法:挖掘Oncomine数据库中NAMPT相关数据,对其在不同类型肿瘤及胶质瘤中的表达情况进行荟萃分析,通过中国脑胶质瘤基因组图谱计划( Chinese Glioma Genome Atlas, CGGA)数据库分析其表达水平与胶质瘤患者预后的关系.通过GO和Pathway数据库对NAMPT基因进行功能注释,包括细胞组分、生物学过程、分子功能及相关信号通路.结果:Oncomine数据库中共收集426 项关于NAMPT在不同肿瘤类型的研究结果,其中有统计学差异的研究共43 项,NAMPT表达增高的研究有24 项,表达降低的研究有19 项;胶质瘤中NAMPT高表达的研究有8项,无低表达的研究.与对照组相比,NAMPT在胶质瘤中高表达(P<0. 01).分析CGGA数据库的基因芯片数据发现,在胶质母细胞瘤中,高表达NAMPT的患者预后更差(P<0. 05).通过GO和Pathway注释发现,NAMPT参与NAD代谢补救途径、细胞与细胞信号传导、昼夜节律调控、细胞增殖、RNA聚合酶II转录调控、信号转导等生物学进程.结论:NAMPT在胶质瘤中高表达,且与胶质母细胞瘤预后有关,或可作为判断胶质母细胞瘤预后的指标.“,”Objective: To explore the expression of nicotinamide phosphoribosyltransferase( NAMPT) and the prognostic role it plays in glioma, and to perform Gene Ontology (GO) and Pathway annotation. Methods: The expression of NAMPT in several kinds of tumors were retrieved from the Oncomine database, and the prognostic data of NAMPT in glioma were re-trieved from Chinese Glioma Genome Atlas (CGGA) database and were analyzed. Biological processes, molecular functions, cellular component and related pathways were analyzed for NAMPT in glioma by GO and Pathway. Results: Totally, 426 studies on expression of NAMPT in different types of tumors were identified in the Oncomine database. The NAMPT expres- sion was significantly different in tumors of 43 studies, and were up-regulated in 24 (8 in glioma) and down-regulated in 19 studies ( none in glioma). NAMPT expression in glioma was higher than that in normal tissue (P<0. 05). Data from CGGA showed that glioblastoma patients with high expression of NAMPT tended to have a worse overall survival (P<0. 05); according to GO and Pathway annotation, NAMPT is in-volved in biological processes including NAD biosynthesis via nicotinamide riboside salvage pathway, cell-cell signaling, cir-cadian rhythm, cell proliferation, RNA polymerase II regulation, and signal transduction etc. Conclusion: NAMPT is highly expressed in glioma, and its expression is associated with the prognosis of glioblastoma, which may be a promising prognostic indicator for glioblastoma.
