目的 研究大蒜素对恶性疟原虫体外生长活性的影响。方法 用新鲜配制或储存的疟疾完全培养基大蒜素稀释液在不同浓度 (10 0 μg/mL~ 5 μg/mL)处理恶性疟原虫 ,实验过程中在 12h ,2 4h换液或者不换液。分别作用 12h ,2 4h和 48h后 ,检测虫体感染率的变化分析药物的作用效果。同时以大蒜素处理正常红细胞观察对虫体宿主细胞的影响。结果 以新鲜配制的大蒜素处理疟原虫时 ,作用 12h后 5 0 μg/mL的浓度即可清除虫体 ,当作用 48h后在低至10 μg/mL的浓度也可杀灭原虫。而使用储存大蒜素换液或者新鲜配制的大蒜素不换液则需要较高的浓度。大蒜素处理正常红细胞时 ,在高于 5 0 μg/mL的浓度对宿主红细胞有破坏作用 ,而该浓度下虫体生长可被有效抑制。 结论 大蒜素在体外具有强烈的抗疟作用。 3 0 μg/mL是可以有效抑制疟原虫生长但对宿主红细胞没有损伤的适宜浓度。
Objective To study the effect of allicin on the growth activity of Plasmodium falciparum in vitro. Methods P. falciparum was treated with various concentrations (100 μg/mL to 5 μg/mL) of malarial complete medium prepared or stored in freshly prepared or malarial dilution medium. During the experiment, the samples were changed or not changed at 12 h and 24 h. After 12 h, 24 h and 48 h respectively, the effect of the drug was analyzed by examining the change in the infection rate of the worms. At the same time, the effects of allicin on normal erythrocytes were observed. Results When freshly prepared allicin was used to treat Plasmodium falciparum, the concentration of 50 μg/mL after 12 h could remove the parasite, and it could also kill the protozoan at a concentration of as low as 10 μg/mL after 48 h. The use of stored allicin or freshly prepared allicin does not require a higher concentration. When allicin is used to treat normal red blood cells, it has a destructive effect on the host red blood cells at a concentration higher than 50 μg/mL, and the growth of the parasites at this concentration can be effectively inhibited. Conclusion Allicin has a strong antimalarial effect in vitro. 30 μg/mL is an appropriate concentration that can effectively inhibit the growth of Plasmodium but does not damage the host red blood cells.
