目的 观察小鼠感染泡球蚴后其体液免疫的动态变化。 方法 BALB/c小鼠感染泡球蚴后 ,分别于 2、4、8、1 2、1 6、2 0及 2 5wk(IgG含量达高峰 )取脾脏制备淋巴细胞悬液体外培养 ,分别以多房棘球蚴抗原 (EmAg)、伴刀豆球蛋白A (ConA)刺激诱生可溶性白细胞介素 2受体 (IL 2R)、肿瘤坏死因子α(TNF α)及白细胞介素 1 (IL 1 ) ;以植物凝集素(PHA)刺激诱生干扰素γ(IFN γ)。检测培养上清中IL 2R、TNF α、IL 1及IFN γ含量。各组均设RPMI1 6 40培养液平行对照。检测血清中一氧化氮 (NO)及特异性免疫球蛋白G(IgG)亚类水平。 结果 小鼠感染泡球蚴 1 6wk后NO水平明显升高 ,IgG、IgG1和IgG3水平升高 ,IgG2a及IgG2b呈低水平。感染后的前 1 2wk ,脾淋巴细胞以分泌IL 2R和TNF α为主 ,1 2wk后以IFN γ为主、1 6wk后以IL 1为主。 结论 小鼠感染泡球蚴后的前 8wk呈Th1反应。感染后期Th2反应逐渐增强
Objective To observe the dynamic changes of humoral immunity in mice after infection of Cysticercus cellulosae. Methods BALB / c mice were infected with metacercariae, and splenocytes were prepared in vitro at 2, 4, 8, 1, 2, 12, 20 and 25wk respectively (peak IgG content) The activity of soluble interleukin 2 receptor (IL 2R), tumor necrosis factor α (TNF α) and interleukin 1 (IL 1) were induced by the presence of EmAg and Con A, ; Induced by phytohemagglutinin (PHA) induced interferon gamma (IFN gamma). The contents of IL 2R, TNF α, IL 1 and IFN γ in culture supernatant were detected. Each group were set up RPMI1 6 40 culture medium parallel control. Serum levels of nitric oxide (NO) and specific immunoglobulin G (IgG) were measured. Results The levels of NO, IgG, IgG1 and IgG3 in mice infected with 1 (superscript th) week after infection of Echinococcus granulosus were significantly increased while the levels of IgG2a and IgG2b were low. In the first 12 weeks after infection, spleen lymphocytes mainly secreted IL-2R and TNF-α, mainly IFNγ after 12 weeks and mainly IL-6 after 16 weeks. Conclusions The mice were infected with metacercariae before 8wk was Th1 response. Th2 response gradually increased after infection
