目的:观察表没食子儿茶素没食子酸酯(EGCG)对人肾小管上皮细胞(HK-2)结缔组织生长因子(CTGF)mRNA和蛋白表达的影响并探讨其可能的机制。方法:体外培养HK-2细胞,EGCG分别以25μmol/L、50μmol/L、100μmol/L作用于体外培养的HK-2细胞48h。用ELISA法和RT-PCR检测HK-2细胞CTGF蛋白含量及mRNA表达水平。结果:与对照组比较,EGCG组HK-2细胞CTGF表达显著下降(P<0.01),且呈浓度依赖关系。结论:EGCG能抑制HK-2细胞CTGF的表达。
Objective: To investigate the effect of epigallocatechin-3-gallate (EGCG) on mRNA and protein expression of connective tissue growth factor (CTGF) in human renal tubular epithelial cells (HK-2) and to explore its possible mechanism. Methods: HK-2 cells were cultured in vitro. EGCG cells were treated with 25μmol / L, 50μmol / L and 100μmol / L in vitro for 48h. The expression of CTGF protein and mRNA in HK-2 cells were detected by ELISA and RT-PCR. Results: Compared with the control group, the CTGF expression of HK-2 cells in EGCG group was significantly decreased (P <0.01), and in a concentration-dependent manner. Conclusion: EGCG can inhibit the expression of CTGF in HK-2 cells.
