黑素瘤缺乏因子2(absentinmelanoma 2,AIM2)是一种细胞质的蛋白质,能够通过PYD(pyrin domain)结构域与接头蛋白( apoptosis-associated speck-like protein containing a CARD,ASC)相互作用。一旦感知到了胞质dsDNA之后,AIM2就会募集到ASC产生出炎症复合体,进而激活了caspase-l,促进炎性细胞因子成熟与分泌。 AIM2还能够感受到胞质与核内dsDNA所发出的不同天然免疫应答,诱导生成干扰素-β和炎性细胞因子。研究表明,AIM2是一种肿瘤抑制因子,因为在许多恶性肿瘤中,如黑色素瘤、结直肠癌和前列腺肿瘤,AIM2表达存在降低或缺失;同时研究表明结直肠癌患者肿瘤细胞中AIM2表达缺失与其预后不良密切相关,但研究人员并不清楚AIM2如何影响肿瘤的发生。最近一些团队研究了AIM2在结直肠癌发生发展中的作用,本文就对此进行总结。“,”Absent in melanoma 2 (AIM2) is a cytoplasmic protein,can through the PYD (pyrin domain) and the adaptor protein ( ASC) interaction.Once the perceived cytoplasmic dsDNA,AIM2 will be raised to ASC produce inflammation complex,activation of Caspase-l,and promote the inflammatory cytokines in the matration and secretion.AIM 2 also can feeld iferent natural immunere-sponse by cytoplasmic and nuclear dsDNA, induced by cytokines and inflammatory produce INF-β.Research also suggests that AIM2 is a tumor suppressor because its expression is turned down or off in many melanomas, colorectal cancers, and prostate tumors;lack of AIM2 expression in tumor cells is closely associated with poor survival in colorectal cancer patients.However, re-searchers don’ t know precisely how AIM2 affects tumorigenesis, so some teams investigated its role on the development of colorectal cancer.This review will summarize this.
