目的探讨变应性鼻炎(AR)大鼠鼻黏膜Fas的表达和雷公藤甲素对他们的干预作用,为雷公藤甲素治疗变应性鼻炎提供初步理论依据。方法选健康SD大鼠40只,雌雄不限,随机分为卵清蛋白(OVA)致敏组,雷公藤(TP)处理组,地塞米松(DM)处理组和生理盐水(SC)对照组,每组10只,用卵清蛋白腹腔注射致敏,继之鼻局部激发建立AR大鼠模型,TP处理组、DM处理组和SC对照组分别用TP、DM和SC在激发前30m in腹腔注射和鼻腔滴入。取各组大鼠鼻黏膜,行Fas蛋白免疫组织化学染色,图像分析软件分析阳性区的灰度值。实验数据采用统计软件包SPSS13.0,统计学分析采用单因素方差分析并继以最小显著差(LSD)t检验比较组间差异。结果变应性鼻炎大鼠鼻黏膜Fas主要表达于上皮、腺体、平滑肌及血管周围的炎性细胞中;TP处理组和DM处理组Fas的表达水平显著高于OVA组(P<0.01),TP处理组和DM处理组间表达水平比较无显著性差异(P>0.05)。结论 TB能够促进大鼠鼻黏膜组织Fas的表达、且与DM比较无显著差异,这可能是其药理作用机制之一。
Objective To investigate the expression of Fas in the nasal mucosa of allergic rhinitis (AR) rats and the effect of triptolide on them, and to provide a preliminary theoretical basis for triptolide treatment of allergic rhinitis. Methods Forty healthy SD rats were randomly divided into sensitized group of ovalbumin (OVA), tripterygium (TP), dexamethasone (DM) and normal saline (SC) , 10 rats in each group were sensitized by intraperitoneal injection of ovalbumin, followed by local nasal stimulation to establish an AR rat model. TP, DM and SC control groups were respectively treated with TP, DM and SC for 30 minutes before challenge Injections and nasal drops. The nasal mucosa of rats in each group were taken for immunohistochemical staining of Fas protein and the gray value of positive area was analyzed by image analysis software. The experimental data using statistical package SPSS13.0, statistical analysis using one-way ANOVA followed by least significant difference (LSD) t test to compare differences between groups. Results Fas in nasal mucosa of allergic rhinitis was mainly expressed in epithelial, glandular, smooth muscle and perivascular inflammatory cells. The expression of Fas in TP group and DM group was significantly higher than that in OVA group (P <0.01) There was no significant difference between TP group and DM group (P> 0.05). Conclusion TB can promote the expression of Fas in nasal mucosa of rats, and there is no significant difference with DM, which may be one of its pharmacological mechanisms.
