目的采用HPLC法测定墨旱莲配方颗粒中原儿茶酸、原儿茶醛的含量。方法采用正交试验进行提取方法的研究。采用Agilent Zorbax Eclipse XDB-C18柱(150 mm×4.6 mm,5μm),甲醇-0.1%磷酸水溶液(10∶90)为流动相,流速1 mL·min-1,检测波长280 nm,柱温为35℃。结果采用20倍量75%甲醇,超声提取1 h可较好地提取墨旱莲配方颗粒中的原儿茶酸与原儿茶醛。原儿茶酸与原儿茶醛的线性范围分别为33.84~112.80(r=0.9999)、3.328~16.64μg·mL-1(r=0.9999),平均加样回收率分别为99.50%、100.83%,RSD分别为0.88%、1.53%(n=6)。结论所用方法结果准确可靠、重复性好,为控制墨旱莲配方颗粒的质量提供科学依据。
OBJECTIVE To determine the contents of protocatechuic acid and protocatechuic aldehyde in the formula particles of Eclallycin by HPLC. Methods The orthogonal experiment was used to study the extraction method. The mobile phase consisted of Agilent Zorbax Eclipse XDB-C18 column (150 mm × 4.6 mm, 5 μm) and methanol-0.1% phosphoric acid solution (10:90) at a flow rate of 1 mL · min-1 with a detection wavelength of 280 nm and a column temperature of 35 ℃. Results The extraction of protocatechuic acid and protocatechuic aldehyde in Eclipta formula granules by ultrasonic extraction for 20 h with 20 times the amount of 75% methanol. The linear ranges of protocatechuic acid and protocatechuic aldehyde were 33.84-112.80 (r = 0.9999) and 3.328-16.64μg · mL-1 (r = 0.9999) respectively. The average recoveries of protocatechuic acid and protocatechuic aldehyde were 99.50% and 100.83% RSD were 0.88%, 1.53% (n = 6). Conclusion The results of the method used are accurate and reproducible, and provide a scientific basis for controlling the quality of Eclipta formula particles.
