目的:研究热休克处理小鼠肥大细胞瘤细胞P815对细胞内Qa-1表达影响。方法:将体外培养的小鼠肥大细胞瘤细胞P815分为对照组及热休克组(3个温度),共4组:对照组细胞置于37℃的水浴箱中1 h,而后将细胞正常培养4 h;热休克组细胞分别置于41℃、42℃和43℃的水浴箱中1 h,然后恢复至37℃。RT-PCR及流式细胞技术检测分析细胞内Qa-1 mRNA及蛋白水平。结果:热休克可引起小鼠肥大细胞瘤细胞P815胞内Qa-1的mRNA和蛋白水平明显升高,与对照组比较,差异均有统计学意义(均P<0.05),其中43℃热休克处理对Qa-1表达的诱导作用最强。结论:热休克可引起小鼠肥大细胞瘤细胞P815 Qa-1表达上调。
OBJECTIVE: To study the effect of heat shock on the expression of Qa-1 in mouse mast cell tumor cell P815. Methods: P815 cells cultured in vitro were divided into control group and heat shock group (3 temperatures), a total of 4 groups: the control group cells were placed in 37 ℃ water bath for 1 h, and then the cells were normal culture 4 h; heat shock group cells were placed in 41 ℃, 42 ℃ and 43 ℃ water bath for 1 h, then returned to 37 ℃. RT-PCR and flow cytometry analysis of Qa-1 mRNA and protein levels in cells. Results: Heat shock induced a significant increase of intracellular Qa-1 mRNA and protein levels in P815 cells, compared with the control group (all P <0.05). Heat shock at 43 ℃ The treatment of Qa-1 expression of the strongest induction. Conclusion: Heat shock can up-regulate the expression of P815 Qa-1 in mouse mast cell.
