抗菌肽是一类具有抗菌活性的多肽类物质。利用农杆菌介导法将中国对虾抗菌肽基因np3和np5分别导入粳稻品种爱知旭中,PCR检测显示,T0代转np3基因和np5基因水稻植株的阳性率分别为36%和45%。RT-PCR检测T1代转基因水稻,结果表明抗菌肽基因在RNA水平上得到表达,而抗菌肽的表达对农艺性状未产生显著影响。用白叶枯病菌小种CR4分别对4个T1代转np3基因和np5基因株系进行接种,所有转基因株系均表现出比对照更高水平的抗性。为了验证抗菌肽基因的广谱抗性,进一步检测了np5转基因株系对3个菌株JS97-2、ZHE173和OS-225的抗性,发现中国对虾抗菌肽基因np5对白叶枯病菌可能具有较广谱的抗性。
Antibacterial peptides are a class of peptides with antibacterial activity. The Agrobacterium tumefaciens-mediated antimicrobial peptide genes np3 and np5 were introduced into Aichi Asakawa, respectively. The results of PCR showed that the positive rates of Tp generation transgenic rice and np5 transgenic rice were 36% and 45% respectively. The results of RT-PCR analysis showed that the antimicrobial peptide gene was expressed at the RNA level, while the expression of the antimicrobial peptide had no significant effect on the agronomic traits. The four T1 generation transgenic np3 and np5 transgenic lines were inoculated with the bacterial blight races CR4, respectively, and all the transgenic lines showed a higher level of resistance than the control. In order to verify the broad-spectrum resistance of the antimicrobial peptide gene, the resistance of the np5 transgenic lines to three strains JS97-2, ZHE173 and OS-225 was further tested and it was found that the np5 antimicrobial peptide gene of P. shrimp may have a wide range of bacterial leaf blight Spectral resistance.
