荞麦发酵物对2型糖尿病db/db小鼠肾VEGF、MMP-9表达的影响

目的观察荞麦发酵物(Fermented Buckwheat Extract,FBE)对2型糖尿病db/db小鼠肾组织血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)、基质金属蛋白酶-9(Matrix Metalloproteinase-9,MMP-9)表达的影响。方法30只8周龄雄性db/db小鼠随机分为FBE高、低剂量组和模型组,10只同周龄同源雄性db/m小鼠作为正常对照组,FBE高低剂量组分别灌胃FBE 100 mg/(kg·d)、50 mg/(kg·d),正常与模型组灌胃等量纯水,连续给药8周。血糖仪测定0、4、8周随机血糖;免疫组化和Western blot法检测肾组织VEGF和MMP-9蛋白表达位置及表达水平。结果模型组小鼠随机血糖明显高于正常对照组(P<0.001),高低剂量FBE治疗组小鼠的随机血糖值明显低于模型组(P=0.009),并且给药8周血糖值降低幅度更大;与正常组相比,模型组小鼠肾脏VEGF蛋白的表达水平明显升高(P<0.001),高、低剂量FBE均能下调VEGF的表达(P均<0.001);MMP-9蛋白表达水平在模型小鼠肾脏明显下降(P<0.001),FBE高、低剂量均可上调小鼠肾脏MMP-9的表达(P均<0.001)。结论 FBE可降低db/db小鼠随机血糖,对db/db小鼠肾组织VEGF蛋白表达有下调作用,对MMP-9蛋白表达有上调作用。 Objective To observe the effect of Fermented Buckwheat Extract (FBE) on the expression of vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9) in renal tissue of type 2 diabetic db / ) Expression. Methods Thirty male 8-week-old db / db mice were randomly divided into FBE high and low dose groups and model group. Ten same-week-old male db / m mice were used as normal control group and FBE high and low dose groups FBE 100 mg / (kg · d), 50 mg / (kg · d). The normal group and the model group were given the same amount of pure water for 8 weeks. Blood glucose was measured at 0,4,8 weeks random blood glucose; immunohistochemistry and Western blot detection of renal tissue VEGF and MMP-9 protein expression and expression levels. Results The blood glucose of model mice was significantly higher than that of normal mice (P <0.001). The high and low dose FBE mice were significantly lower than the model mice (P = 0.009) (P <0.001). Compared with normal group, the expression of VEGF protein in kidney of model group was significantly increased (P <0.001), the expression of VEGF was down-regulated in high and low dose FBE mice (all P <0.001) The expression of MMP-9 in renal tissues of model mice decreased significantly (P <0.001), and the expression of MMP-9 in renal tissues increased in FBE high and low dose groups (all P <0.001). Conclusion FBE can reduce the random blood glucose of db / db mice, down-regulate the expression of VEGF protein in the kidney of db / db mice and up-regulate the expression of MMP-9.
