目的:考察0.1%~0.4%蒽酮-硫酸试液对白及溶胶中多糖含量测定结果的影响,筛选最佳染色剂浓度与用量,提高检出效率。方法:用无水葡萄糖为对照品,0.1%~0.4%蒽酮-硫酸试液为染色剂分组染色白及溶胶供试液,分光光度法测定含量,测定波长620 nm,比较各组吸光度、含量测定结果、回收率。结果:4组染色剂中,0.2%蒽酮-硫酸试液用量5~6 ml时吸光度、含量测定、回收率均优于其他组,回收率为95.6%,RSD=1.61%(n=6),检出效果依次为:0.2%组>0.1%组>0.3%组>0.4%组。结论:染色剂浓度对测定结果有较大影响,使用0.2%浓度染色剂检出率高,重复性好,适用于白及溶胶的含量测定。
OBJECTIVE: To investigate the effect of 0.1% ~ 0.4% anthrone-sulfuric acid test solution on the determination of polysaccharide in white and sol, screen the optimal concentration and dosage of colorant, and improve the detection efficiency. Methods: Anhydrous glucose was used as reference substance. 0.1% -0.4% anthrone-sulfuric acid solution was used to dye white and sol test solution. The content was determined by spectrophotometry. The wavelength was set at 620 nm. The absorbance and content Determination results, recovery rate. Results: The absorbency, the determination and the recovery of 0.2% anthrone-sulfuric acid solution in 5 ~ 6 ml were the best among the four groups. The recoveries were 95.6%, RSD = 1.61% (n = 6) The detection results were as follows: 0.2% group> 0.1% group> 0.3% group> 0.4% group. Conclusion: The concentration of colorant has great influence on the determination results. The detection rate of 0.2% colorant is high and the repeatability is good. It is suitable for the determination of white and sol.
