目的探讨低剂量棉酚12mg/(kg·d)、甲基睾丸酮20mg/(kg·d)和炔雌醇100μg/(kg·d)联合用药24周,能否影响大鼠生精干细胞的功能。方法采用生精细胞原位凋亡、生精干细胞移植和生殖能力自主恢复实验。结果联合用药24周后,在大鼠睾丸组织切片中发现了凋亡信号阳性细胞,凋亡信号阳性细胞主要是精母细胞和精子细胞,没有发现精原细胞出现凋亡阳性信号。当把用药组大鼠的生精干细胞移植进入受体裸鼠的曲细精管内2~3个月后,在受体裸鼠的曲细精管中观察到了来自供体大鼠的长型精子细胞。停止用药6周后,用药组大鼠的生殖能力与对照组相比完全恢复。由恢复生殖能力的用药组大鼠产生的F1代仔鼠在经过一系列的检测后被证明为身体机能和智力水平正常。结论此联合用药在24周内没有影响到用药大鼠生精干细胞的生存和分化功能。
Objective To investigate whether the combination of low dose gossypol (12 mg / (kg · d), methyltestosterone 20 mg / (kg · d)) and ethinylestradiol 100 μg / (kg · d) for 24 weeks can affect the function of rat spermatogenic stem cells . Methods Spermatogenic cells in situ apoptosis, sperm stem cell transplantation and reproductive capacity of self-recovery experiments. Results After 24 weeks of combination treatment, apoptosis-positive cells were found in the testicular tissue sections of rats. The apoptotic signal-positive cells were mainly spermatocytes and spermatids. No apoptotic signal was found in the spermatogonia. When the spermatogenic stem cells of the drug-treated rats were transplanted into the seminiferous tubules of the recipient nude mice for 2 to 3 months, long spermatozoa from donor rats were observed in the seminiferous tubules of the recipient nude mice cell. After 6 weeks, the reproductive capacity of the rats in the treatment group recovered completely compared with the control group. Fl generation of F1 mice from fertility-treated rats proved to be normal in both physical and mental status after a series of tests. Conclusion This combination did not affect the survival and differentiation of spermatogonial stem cells in 24 weeks.
