目的观察低浓度三氧化二砷(As2O3)在不同氧环境下对肺癌A549细胞增殖抑制、细胞周期阻滞、细胞凋亡的诱导作用。方法分别在常氧(21%O2)、低氧(5%O2)、厌氧(0%O2)环境利用0μmol/L As2O3(空白对照组)、1μmol/L As2O3、1μmol/L VP-16体外诱导培养肺癌A549细胞24h,通过甲基噻唑基四唑(methyl thiazolyl tetrazolium,MTT)法测细胞增殖抑制率,通过碘化丙啶(propidium iodide,PI)染色、流式细胞仪检测细胞周期,通过AnnexinⅤ/PI双染色法检测细胞凋亡。结果 1μmol/LAs2O3组A549细胞增殖抑制率及G0/G1期细胞比例随缺氧加重而增加;厌氧环境下该组细胞增殖抑制率及G0/G1期细胞比例均高于空白对照组(P<0.05),但其细胞凋亡率较空白对照组降低(P<0.05);与3种相应氧环境下VP-16组相比较,As2O3组A549细胞增殖抑制率、G0/G1期细胞比例、G2/M期细胞比例、细胞凋亡率均减少(P<0.05)。结论 1μmol/LAs2O3对A549细胞无明显诱导凋亡作用,厌氧环境下该剂量As2O3可以通过G1期阻滞抑制A549细胞增殖。
Objective To observe the effect of low concentration of arsenic trioxide (As2O3) on the proliferation inhibition, cell cycle arrest and apoptosis of lung cancer A549 cells in different oxygen environment. Methods 0μmol / L As2O3 (blank control group), 1μmol / L As2O3 and 1μmol / L VP-16 were used in vitro in the environments of normoxic (21% O2), hypoxic (5% O2) and anaerobic (0% The A549 cells were induced to culture for 24 hours. The inhibitory rate of cell proliferation was determined by methyl thiazolyl tetrazolium (MTT) assay. The cell cycle was detected by propidium iodide (PI) staining and flow cytometry. Annexin Ⅴ / PI double staining was used to detect apoptosis. Results The inhibition rate of A549 cells and the proportion of G0 / G1 phase cells in 1μmol / L As2O3 group increased with hypoxia. The cell proliferation inhibition rate and the proportion of G0 / G1 phase cells in anaerobic environment were higher than those in blank control group (P < 0.05), but the apoptosis rate was lower than that of the blank control group (P <0.05). Compared with the VP-16 group under the three oxygen conditions, the inhibition rate of A549 cells, the proportion of G0 / G1 phase cells, / M phase cell ratio, apoptosis rate were decreased (P <0.05). Conclusion 1μmol / L As2O3 can not induce apoptosis of A549 cells. As2O3 can inhibit the proliferation of A549 cells through G1 phase arrest in anaerobic environment.
