目的:研究益气软坚散结法对甲状腺功能减退大鼠心肌TRα1mRNA表达的影响。方法:(1)将Wist-ar大鼠随机分为正常组、模型组、L-T4组、益气软坚散结组(自拟芪贝复元饮)4组,每组30只。(2)正常组:普通饲料,双蒸水;模型组:低碘饲料,双蒸水。(3)酶联免疫吸附测定法(Elisa法)测大鼠血清促甲状腺激素(TSH),放射免疫分析法(RIA)测血清总T3(TT3),总T4(TT4)。实时荧光定量PCR(Real-time PCR法)检测大鼠心肌TRα1mRNA的表达。砷铈分光光度法测定尿碘含量。光镜下观察大鼠心肌心肌形态。心电图机记录大鼠心电变化。结果:56天时予处理因素的L-T4组和芪贝复元饮组大鼠血清TT3值均升高(P<0.01),二组之间比较无差异;血清TT4值均升高(P<0.001),二组之间比较无差异;TSH值明显降低(P<0.01),其中较L-T4组血清TSH值降低明显。心电图描记发现芪贝复元饮组与L-T4组比较有差异(P<0.05),心率及低电压表现有恢复;芪贝复元饮组心肌TRα1mRNA的表达较L-T4组高0.81倍(P<0.05);心肌细胞形态观察发现芪贝复元饮组较L-T4组恢复的明显。结论:与L-T4比较益气软坚散结法能使甲减心肌损害得到恢复,其作用机制与TRα1mRNA在心肌细胞上的表达有关。
Objective: To study the effects of Yiqi Ruanjian Sanjie on myocardial TRα1 mRNA expression in hypothyroid rats. Methods: (1) Wist-ar rats were randomly divided into 4 groups: normal group, model group, L-T4 group and Yiqi Ruanjian Sanjie group (self-administered Qibei Fuyuan Decoction), with 30 rats in each group. (2) normal group: ordinary feed, double distilled water; model group: low iodine feed, double distilled water. (3) Serum thyrotropin (TSH) was measured by Elisa method, total T3 (TT3) and total T4 (TT4) were measured by radioimmunoassay (RIA). Real-time fluorescence quantitative PCR (Real-time PCR) was used to detect the expression of TRα1 mRNA in rat myocardium. Determination of urinary iodine content with arsenic and cerium spectrophotometry. Myocardial morphology was observed under light microscope. ECG ECG changes in rats. Results: The serum TT3 of L-T4 group and Qibei Fuyuan Decoction group were all increased at 56 days (P <0.01), no difference was found between the two groups (P <0.001) ), No difference between the two groups; TSH value was significantly lower (P <0.01), which was significantly lower than the L-T4 serum TSH value. Electrocardiogram showed that Qibei Fuyuan Decoction had a significant difference compared with L-T4 group (P <0.05), heart rate and low voltage showed recovery, TRibe mRNA expression in Qibei Fuyuan Decoction group was 0.81 times higher than L-T4 group (P < 0.05). Myocardial morphological observation showed that Qibei Fuyuan Decoction recovered more obviously than L-T4 group. CONCLUSION: Compared with L-T4, Yiqi Ruanjian Sanjie can restore the myocardial damage caused by hypothyroidism and its mechanism is related to the expression of TRα1 mRNA in cardiomyocytes.
