目的 观察并比较不同亚型金属硫蛋白 (MT)与心肌血管内皮细胞以及MT与缺血再灌注损伤后心肌血管内皮细胞的特异结合。方法 以胶体金为示踪物 ,采用离体心脏灌流模型 ,电镜下观察心肌血管内皮细胞对MT的特异结合。结果 不同亚型MT(MT 1,MT 2 )与心肌血管内皮细胞特异结合无明显差异。心脏缺血 30min和 6 0min后心肌血管内皮细胞仍能特异结合MT ,在心脏缺血 30min时 ,心肌血管内皮细胞特异结合MT的量与正常有氧灌注相比无明显差异 ,心脏缺血 6 0min时 ,心肌血管内皮细胞特异结合MT的量明显减少。结论 为进一步研究该蛋白生理功能及其应用提供重要依据。
Objective To observe and compare the specific binding of different subtypes of metallothionein (MT) to myocardial vascular endothelial cells (MTs) and MT to myocardial vascular endothelial cells after ischemia-reperfusion injury. Methods Colloidal gold was used as a tracer. The isolated cardiac perfusion model was used to observe the specific binding of MT to endothelial cells of myocardium under electron microscope. Results There was no significant difference between the specific subtypes of MT (MT 1, MT 2) and myocardial vascular endothelial cells. Myocardial endothelial cells could still specifically bind to MT at 30min and 60min after ischemia, and there was no significant difference in the amount of MT specific binding to myocardial vascular endothelial cells between 30min and 60min , The amount of specific binding of MT to myocardial vascular endothelial cells was significantly reduced. Conclusion provides an important basis for further study of the physiological function of the protein and its application.
