目的 :观察新型免疫抑制剂雷帕霉素 (Rapa mycin ,RPM)对异种单向混合淋巴细胞培养 (one wayXMLC)的免疫抑制作用。方法 :①无菌采集小鼠脾淋巴细胞 ,经丝裂毒素处理 ,作刺激细胞 ;②取健康成人外周血淋巴细胞(hPBL) ,作反应细胞 ;③异种单向混合淋巴细胞培养 ;④实验分对照组 (不用药 )、RPM组 ,实验组 (用药组 )又分不同药物浓度组。结果 :①小鼠脾淋巴细胞明显刺激人外周血淋巴细胞 (hPBL)增殖 ,3H TdR掺入值 (cpm)显著增高 (P <0 .0 5 ) ;CD4 、CD8、IgG、IgM细胞含量明显升高 ,有显著差异 (P <0 .0 5 )。②各实验组的cpm值下降 ,RPM的IC50 (5 0 %有效抑制浓度 )≈ 1.5nmol L。测定各实验组的CD4 ,CD8,IgG ,IgM细胞含量下降 ,药物的IC50 与上相似。结论 :①人→小鼠单向混合淋巴细胞培养 ,具有明显细胞增殖反应。②RPM对异种单向混合淋巴细胞培养具有明显的免疫抑制效果 ,其作用呈剂量依赖性。
OBJECTIVE: To observe the immunosuppressive effect of Rapamycin (RPM), a novel immunosuppressive agent, on one-way XMLC. Methods: ① Splenic lymphocytes from mice were collected aseptically and treated with mitomycin C for stimulating cells. ② Healthy adult peripheral blood lymphocytes (hPBL) were used as reaction cells. ③ Heterogenous one-way mixed lymphocyte culture Control group (without medication), RPM group, experimental group (medication group) were divided into different drug concentration groups. Results: ① The splenic lymphocytes of mice significantly stimulated the proliferation of human peripheral blood lymphocytes (hPBL), and the incorporation of 3H TdR (cpm) was significantly increased (P <0.05). The contents of CD4, CD8, IgG and IgM were significantly increased High, there are significant differences (P <0. 05). ② The cpm value of each experimental group decreased, and the IC50 of RPM (50% effective inhibitory concentration) ≈1.5nmol L. The contents of CD4, CD8, IgG and IgM in each experimental group were determined to be decreased, and the IC50 of the drug was similar to the above. Conclusion: ① human → mouse unidirectional mixed lymphocyte culture, with significant cell proliferation response. ②RPM had obvious immunosuppressive effect on the heterogeneous unidirectional mixed lymphocyte culture in a dose-dependent manner.
