目的运用逆转录聚合酶链反应(RT-PCR)技术研究青蒿琥酯对肿瘤细胞DNA损伤修复基因表达的影响。方法用青蒿琥酯处理K562细胞72 h,倒置光显微镜下观察细胞形态变化,Trizol法提取总RNA,RT-PCR检测DNA依赖性蛋白激酶(DNA-PK)的表达变化。结果倒置光显微镜观察到青蒿琥酯处理的K562细胞出现不同程度的皱缩,核分裂相减少,细胞密度下降,漂浮细胞增多。RT-PCR检测结果显示DNA-PK表达下调。结论青蒿琥酯可通过下调K562细胞DNA-PK表达,诱导K562细胞凋亡发挥其抗癌作用。
Objective To study the effect of artesunate on DNA damage repair gene expression in tumor cells by reverse transcription-polymerase chain reaction (RT-PCR). Methods K562 cells were treated with artesunate for 72 h. The morphological changes were observed under inverted light microscope. Total RNA was extracted by Trizol method. The expression of DNA-PK was detected by RT-PCR. Results Inverted light microscopy showed that artesunate-treated K562 cells showed different degrees of shrinkage, decreased mitotic phase, decreased cell density and increased floating cells. RT-PCR results showed that DNA-PK expression was down-regulated. Conclusion Artesunate can exert its anticancer effect by down-regulating DNA-PK expression in K562 cells and inducing apoptosis in K562 cells.
