目的:研究大孔树脂对桑黄总三萜的吸附特性,优化其对总三萜分离纯化工艺。方法:用分光光度计法测定总三萜含量,通过静态试验确定最适合纯化总三萜成分的树脂型号,研究D101树脂对桑黄总三萜的纯化工艺。结果:D101树脂对桑黄总三萜有良好的纯化性能,工艺条件为:上样液p H值为6,上样液质量浓度为1.0mg/m L,上样流度为3BV/h,上样体积为4BV,先用3BV蒸馏水、20%乙醇溶液2BV洗去杂质、再用70%乙醇溶液4BV洗脱,流速为3BV/h;经D101树脂纯化后,总三萜纯度由(4.46±0.42)%上升至(22.49±0.75)%。结论:D101树脂可用于桑黄总三萜的富集纯化。
Objective: To study the adsorption properties of macroporous resin on total triterpenoids of Phellinus igniarius and to optimize the separation and purification of total triterpenoids. Methods: The content of total triterpenoids was determined by spectrophotometer. The optimum triterpenoid resin type was determined by static test. The purification of total triterpenoids by D101 resin was studied. Results: The D101 resin had good purification properties for the total triterpenoids of Phellinus linteus. The process conditions were as follows: the p H value of the sample solution was 6, the mass concentration of the sample solution was 1.0 mg / m L, the sample fluidity was 3BV / h, The sample volume was 4BV, first with 3BV distilled water, 20% ethanol solution 2BV wash impurities, and then 70% ethanol solution 4BV elution, the flow rate of 3BV / h; After D101 resin purification, the total triterpenoid purity by the (4.46 ± 0.42)% to (22.49 ± 0.75)%. Conclusion: D101 resin can be used for enrichment and purification of total triterpenoids in Phellinus igniarius.
