目的:建立人血清中3种抗癫痫药物同时用HPLC法测定,为人体内药代动力学研究和药效学评价提供方法基础。方法:取血清0.1ml,以地西泮为内标,用无水乙醚一次提取,水浴40~50℃,氮气吹干,残渣用流动相溶解,进样20μl。结果:平均回收率(n=3)苯巴比妥99.9%,苯妥英95.9%,卡马西平99.4%。日内差(RSD)均小于1.9%,日间差(RSD)均小于6.1%,线性相关系数在0.995以上。结论:本方法能快速、简便、准确地测出3种抗癫痫药物的血清浓度,有利于临床药代动力学的研究。
OBJECTIVE: To establish three kinds of antiepileptic drugs in human serum by HPLC and to provide a method basis for pharmacokinetic study and pharmacodynamic evaluation in human. Methods: Take serum 0.1ml, take diazepam as internal standard, extract once with anhydrous diethyl ether, and use water bath 40 ~ 50 ℃, dry with nitrogen and dissolve the residue in mobile phase. Results: The average recovery (n = 3) was 99.9% for phenobarbital, 95.9% for phenytoin and 99.4% for carbamazepine. The intraday difference (RSD) was less than 1.9%, the daytime difference (RSD) was less than 6.1%, and the linear correlation coefficient was above 0.995. Conclusion: This method can quickly, easily and accurately measure the serum concentrations of three kinds of anti-epileptic drugs, is conducive to the study of clinical pharmacokinetics.
