目的研究红景天苷(SDS)对百草枯(PQ)中毒大鼠肺纤维化肺组织中基质金属蛋白酶-2(MMP-2)及其组织特异性抑制因子-1(TIMP-1)表达的影响。方法 SD大鼠随机分为对照组,PQ模型组和SDS治疗组。一次性灌胃PQ制备肺纤维化模型,治疗组腹腔注射SDS(10 mg/kg,q12 h);正常对照组腹腔注射等体积NS。PQ模型组和SDS治疗组分别在染毒后相应时间点(1 d,3 d,7 d,14 d,21 d,28 d)麻醉处死大鼠;取其肺组织,进行HE染色;免疫组化和Real-Time PCR检测MMP-2、TIMP-1蛋白与mRNA表达。结果与NS组比较,模型组MMP-2 mRNA及其蛋白、TIMP-1 mRNA及蛋白的表达明显增强(P<0.01);各干预组肺组织表达相对减低(P<0.05)。结论 SDS能降低MMP-2及TIMP-1的mRNA及蛋白的表达,抑制PQ中毒大鼠肺组织的纤维化。
Objective To study the effects of salidroside on the expression of matrix metalloproteinase-2 (MMP-2) and tissue-specific inhibitor of metalloproteinase-1 (TIMP-1) in pulmonary fibrosis of paraquat influences. Methods SD rats were randomly divided into control group, PQ model group and SDS treatment group. Pulmonary fibrosis model was established by intragastric administration of PQ. The treatment group was given intraperitoneal injection of SDS (10 mg / kg, q12 h). The normal control group was injected intraperitoneally with equal volume of NS. The rats in PQ model group and SDS treatment group were anesthetized and killed at the corresponding time point (1 d, 3 d, 7 d, 14 d, 21 d, 28 d) respectively. The lung tissues were taken for HE staining. Real-time PCR was used to detect the expression of MMP-2, TIMP-1 protein and mRNA. Results Compared with NS group, the expression of MMP-2 mRNA and its protein, TIMP-1 mRNA and protein in model group were significantly increased (P <0.01). The expression of lung tissue in each group was significantly decreased (P <0.05). Conclusion SDS can decrease the mRNA and protein expression of MMP-2 and TIMP-1 and inhibit the lung fibrosis in PQ-poisoned rats.
