目的探讨葡萄糖胺对肿瘤坏死因子-α(tumornecrosis factor-α,TNF-α)诱导的人脐带血管内皮细胞(HUVEC)血管细胞黏附分子(vascular cell adhesion molecule,VCAM-1)表达的影响。方法实时逆转录聚合酶链式反应(Real time RT-PCR)和免疫印迹法(Western blot)分别测定葡萄糖胺对VCAM-1的表达情况;Western blot法测定HUVEC核因子-κB(nuclear factor,NF-κB)磷酸化程度。结果葡萄糖胺抑制TNF-α诱导的VCAM-1在HUVEC的表达;NF-κB抑制剂BMS-345541抑制TNF-α诱导的VCAM-1的表达;葡萄糖胺抑制TNF-α诱导的NF-κB的磷酸化。结论葡萄糖胺抑制TNF-α诱导的VCAM-1的表达与抑制NF-κB的活化相关。
Objective To investigate the effect of glucosamine on the expression of vascular cell adhesion molecule (VCAM-1) induced by tumor necrosis factor-α (TNF-α) in human umbilical cord blood endothelial cells (HUVECs). Methods Real-time RT-PCR and Western blot were used to detect the expression of VCAM-1 by glucosamine. Western blot was used to detect the expression of nuclear factor-kappa B (NF-κB) -κB) phosphorylation. Results Glucosamine inhibited the expression of VCAM-1 induced by TNF-α in HUVEC. The NF-κB inhibitor BMS-345541 inhibited the expression of VCAM-1 induced by TNF-α. Glucosamine inhibited the phosphorylation of TNF-α induced NF- The Conclusion Glucosamine inhibits the TNF-α-induced VCAM-1 expression and the inhibition of NF-κB activation.
